Preparation of Biodegradable Microparticles Using Microfluidic Device and Its Applications

Abstract

Microencapsulation is a widely used method for precise delivery of drugs, vaccines, and various functional materials. In order to provide efficient loading and deliver of target materials, it is important to produce carriers that have homogeneous shapes and narrow size distribution. Preparation of microparticles of poly(lactic-co-glycolic acid) (PLGA), a widely used biopolymer, will be presented using microfluidic devices, e.g. geometrically passive T-junction breakup as a high-throughput preparation method for polymer microcarriers. In passive T-junction breakup a polymer solution droplet is split into numerous smaller droplets as it passes a T-junction, and is then polymerized into particles as solvents evaporate. The intrinsic size uniformity, controlled by microfluidic flow and droplet fission, allowed for high-throughput preparation of biopolymer particles at a rate of 8000 particles per second and size distribution of CV < 5%. Prepared microparticles were used as microcarriers for functional materials of drugs and cells. Highly porous microparticles were also prepared successfully using gelatin as a porogen in the T-juntion breakup device. Other research works in our lab will also be briefly introduced regarding micro/nanoscale fabrication and its applications into bioscience.
